In order to characterize and identify pathogenic isolates of this fungus at the molecular level, five symptomatic trees were sampled from farms located in the cantons of: Buena Fe, Mocache, Quevedo, Quinsaloma and Valencia of the province of Los Rios, obtaining 15 isolates from which DNA was extracted and sequences from the ITS (Internal transcribed spacer) and EF1-a (Elongation factor of alpha) regions were used. Sequence sizes were verified by electrophoresis and putative identities of 3 species were established using the Basic local aligment tool (BLAST). Phylogenetic trees constructed using maximum likelihood and nearest neighbor algorithms, with isolates in five cantons, confirmed the presence of Lasiodiplodia theobromae in Quevedo, Diplodia seriata in Quinsaloma and Botryosphaeria dothidea in Valencia, Mocache and Buena Fe, highlighting a polytomy among the latter.

Especies de la familia Botryosphaeriaceae están asociadas a cancros y secamiento de yemas, muerte descendente, pudrición de frutos y pudrición del cuello, síntomas que se han observados en diferentes zonas y que merman la producción de la planta. Con el objetivo de caracterizar e identificar a nivel molecular aislamientos patogénicos de este hongo, se muestrearon cinco árboles sintomáticos seleccionados de fincas localizadas los cantones: Buena Fe, Mocache, Quevedo, Quinsaloma y Valencia de la provincia de Los Ríos, obteniendo 15 aislamientos de los cuales se extrajo ADN y utilizaron secuencias de las regiones de ITS (Internal transcribed spacer) y EF1-a (Factor de elongación de alpha). Los tamaños de secuencia se verificaron mediante electroforesis y se establecieron las identidades putativas de 3 especies utilizando la herramienta de Basic local aligment (BLAST). Los árboles filogenéticos construidos mediante los algoritmos de máxima verosimilitud y vecinos cercanos, con los aislamientos en cinco cantones, confirmaron la presencia de Lasiodiplodia theobromae en Quevedo, Diplodia seriata en Quinsaloma y Botryosphaeria dothidea en Valencia, Mocache y Buena Fe, resaltando una politomía entre estos últimos.Problemas fitossanitários causados por fungos vasculares que afetam as plantações de cacau CCN-51 tornaram-se muito importantes nas últimas décadas, evido aos danos que causam. Espécies da família Botryosphaeriaceae estão associadas a cancros e ressecamento de gemas, morte, podridão dos frutos e podridão do pescoço, sintomas que têm sido observados em diferentes áreas e que reduzem a produção vegetal. Para caracterizar e identificar isolados patogênicos deste fungo a nível molecular, foram amostradas cinco árvores sintomáticas selecionadas em fazendas localizadas nos cantões: Buena Fe, Mocache, Quevedo, Quinsaloma e Valencia na província de Los Ríos, obtendo 15 isolados de do qual o DNA foi extraído e utilizou sequências das regiões ITS (Espaçador transcrito interno) e EF1-a (fator de alongamento alfa). Os tamanhos das sequências foram verificados por eletroforese e as identidades putativas de 3 espécies foram estabelecidas usando a ferramenta de alinhamento local básico (BLAST).
