description The bacterium Ralstonia solanacearum is considered to be highly aggressive, causing disease of moko that leads to the sudden death of Musaceae and other crops. The infection has reached several countries in Latin America, among which the possible introduction of this bacterium to Ecuador from Colombia was reported; country that had little knowledge about this bacterium, which is why it increased the interest of its study to know to which provinces it has dispersed and achieve in future studies the ecology and epidemiology of it. For the isolation of the bacteria, the South Africa Semi-selective Medium (SMSA) was used, which identifies the characteristics of the bacterial colonies and their subsequent purification was carried out through the Nutritive Agar medium. Together, the detection of the bacteria was compared with the DAS-ELISA technique and the microbiological identification system (Biolog Inc.), in which the biochemical components were determined; in addition, the polymerase chain reaction (PCR) was performed to subsequently identify biovars by means of metabolic tests. Were evaluated 80 samples of fruit and pseudostem of different varieties of banana from different provinces of the country that were provided by the Ecuadorian Agency for Quality Assurance of Agro (AGROCALIDAD). The SMSA medium allows detecting colonies of R solanacearum and the microbiological identification system Biolog detected antagonistic bacteria. The DAS-ELISA technique detected R solanacearum and evaluated the inoculum quantity to more than 108UFC/mL. With the PCR technique the bacterium is confirmed and through the positive results it was identified by the degradation of disaccharides (cellobiose, lactose and maltose) and oxidation of polyalcohols (dulcitol, sorbitol and mannitol); with which three types of biovars were obtained: biovar 1 in the province of Guayas, biovar 2 in Manabí and Guayas and biovar 3 in Manabí, Sucumbíos and Napo.
