Sumari:IX ABSTRACT This study evaluated three cryopreservation protocols for high-genetic-merit canine semen to identify the extender that best preserves sperm quality after freeze–thawing. Semen samples were collected from six clinically healthy dogs of various breeds and sizes and processed with three experimental extenders. Microscopic assessments performed after dilution and cryopreservation focused on total motility, normal sperm morphology, and cell viability. The third extender (D3)— a Tris-based formula supplemented with egg yolk and antibiotics—produced significantly higher values (p < 0.05), averaging 70.83 % motility, 75.83 % normal morphology, and 69.5 % viability. These findings support the development of a practical, effective canine semen cryopreservation protocol adapted to Ecuadorian conditions and resources. Its implementation will facilitate the creation of regional germplasm banks, promote genetic diversity conservation in dogs, and strengthen national assisted-reproduction programs.
