Summary:ABSTRACT: Universally, typhoid fever is a health problem caused mainly by poor sanitation that severely affects developing countries. It is essential to know adequate diagnostic techniques for the determination of its etiological agent, Salmonella typhi. The main objective of this study was to validate through a literature review the stool culture versus the Polymerase Chain Reaction in the identification of S. typhi and thus recognize the optimal technique to provide a timely diagnosis. This was a qualitative, descriptive, non-experimental documentary, cross-sectional and retrospective research. The result was a study population of 150 scientific documents dealing with the subject; these documents are located in scientific databases such as PubMed, Scielo, Redalyc, Elseiver, etc., and have been published over a period of 10 years, obtaining a sample of 79 publications related to the subject to be dealt with. In the bibliography consulted, 67% of the authors prefer and recommend the Polymerase Chain Reaction, as opposed to the 33% corresponding to the stool culture. Both techniques present characteristics that make them feasible in 70% (stool culture) and 74% (Polymerase Chain Reaction). In conclusion, both techniques are feasible under the implementation of an adequate protocol, equipment and work area, allowing an adequate specificity and sensitivity, and providing a profitable, suitable and accurate identification of S. typhi.
