Resumo:The present research entitled "In vitro study of the internal anatomy of root canals of the first upper premolar, by means of the dental diaphanization technique", aimed to characterize the internal anatomy of the first upper premolar through the technique of dental diaphanization to determine the number of root canals. The same that consists of transparent the tooth in vitro by means of the application of the chemical substances, the same that intervene in the organic and inorganic components of the tooth. For this, we used 80 first upper premolars as the sample of a universe of 100 teeth (pieces # 14 and # 24), obtained in the Dental Department of the General Teaching Hospital of Riobamba, later they were a diaphanization, following the protocol by Robertson , and the Venturi technique to be able to observe the internal morphology in a three-dimensional way. Giving as results the type of internal anatomy of the first upper premolars of two root canals represented by 65%, as well as the prevalence of the root canals according to the Vertucci classification that the pieces that present the two conduits belong to six types, of which the most important are type IV with 37% followed by type II with 24%, they have an oval shape and an average size of 20.15 mm, that is to say, they vary from 20mm to 21mm.
