Beskrivelse
Summary:The study of the development of pancreatic lipase activity in guinea pigs (Cavia porcellus) from birth to 7 weeks of age; for which the shed of the guinea pig Program was used of the Quinta Experimental Punzara of the National University of Loja located at 2160 m.s.n.m. 22 pregnant females which were fed with commercial equilibrium (PB 18%, 5% fat, 6% crude fiber) will were used, and from which 21 offspring were obtained that were sacrificed on days 0, 7, 14, 21, 28, 35 and 42. The sacrifice was made by cervical dislocation and bled, subsequently the pancreas of each animal under study was extracted, were placed in sterile jars and stored in the freezer. Protein extraction from pancreas samples was performed using the lysis and cold precipitation method; to determine the concentration of soluble protein, the biuret method was carried out and the bovine serum albumin was taken as curve of standard protein. To measure enzymatic activity a commercial Kit (Lipase Activity Colorimetric Assay Kit, BioVision) will was used. The concentration of soluble protein in the pancreas does not show significant differences (P 0,816) with respect to the age of the animal, giving an average of 6,261 mg of protein / mg of sample. The specific activity shows a sharp decrease at 14 days (0,040mU / mg of protein), an increase at 21 days (0,083mU / mg of protein) and 35 to 42 days (0,041mU / mg of protein and 0,092mU / mg of protein) is normalized. The study revealed that the concentration of soluble protein and the specific activity of lipase decrease after weaning, due to the change in diet and stress that animals suffer during this stage.
