Summary:The objective of the study was to establish measures of the reproductive system, observe epithelial-vaginal cells and verify the efficiency of synchronization protocols with reduction in application times. Three experiments were conducted. Experiment 1 identified measurements of ovaries, oviducts, horns and uterine body, cervix and vaginal canal, in addition to the weight of the ovaries of 56 guinea pigs aged 8, 9, 10 and 12 weeks; in experiment two in 69 guinea pigs the vaginal membrane was observed and cytology was used to count and classify the cells of the vaginal epithelium; in experiment three two protocols were applied, Ovsynch and Ovsynch + Progesterone (O+P4); the same analysis of experiment 2 was performed and ultrasound was used for diagnosis of pregnancy. For the results obtained, descriptive statistics and analysis of ANOVA variance and Chi-square test (X2) were performed. In Experiment one: age influenced absolute measurements of ovaries, uterine horns, cervix and vaginal canal being greater (p≤0.05) in guinea pigs of 12 weeks; relative measurements were greater (p≤0.05) in most guinea pig structures of 8 weeks. Experiment two: there was dependence (p<0.001) of the vaginal membrane with the type of cells observed. Experiment three: both protocols showed a higher percentage of parabasal cells (47.85±21.22% and 41.23±24.22%); there was dependence (p<0.001) between the vaginal membrane, cell type and protocol application. The pregnancy percentage showed no difference (p>0.05) between treatments. It is concluded that the age and weights of guinea pigs influenced the absolute and relative measures of the reproductive organs; the vaginal membrane is related to the type of vaginal epithelial cells and the protocols of synchronization, which, despite not being efficient to present oestrus, influenced the presentation of the type of cells and vaginal membrane. Keywords: Ovsynch, guinea pig, oestrus, uterus, vaginal membrane.
