Determinación de la interacción de una celulasa recombinante con la expansina PcExl1 de Pectobacterium carotovorum

The cell wall of plants is made up of several polysaccharides, including cellulose, hemicellulose, and pectin. The cellulose, in turn, possesses crystalline and amorphous regions, which are hydrolyzed by the action of enzymes named as cellulases, endoglucanases and exoglucanases. The bacterium Pectobacterim carotovorum is notorious for its maceration to the cell wall of plants causing soft rot. The ability to destroy plants is due to the expression and secretion of a wide range of hydrolytic enzymes due to the action of the induction system of Gram-negative bacteria, which includes cellulases and expansins. P. carotovorum PC1 FJ890936 was used for the design of primers for amplification of the cellulase gene and to include the cell pelB export site. The PCR product was cloned into the expression vector pET22b-PcExl1 and transformed into Escherichia coli. The molecular weight of each of the clones obtained was 1267 bp. estimating the molecular weights of the expressed protein was performed using SDS-PAGE analysis and two bands of 39 kDa and 32 kDa, respectively. The enzymatic activity was carried out by a reducing sugar release reduction assay. However, cellulase did not show a possible interaction with the expansin PcExl1. La pared celular de las plantas está conformada por varios polisacáridos, entre ellos la celulosa, hemicelulosa, y la pectina. La celulosa a su vez, posee regiones cristalinas y amorfas, que son hidrolizadas por la acción de enzimas nombradas como celulasas, endoglucanasas y exoglucanasas. La bacteria Pectobacterim carotovorum es notoria por su maceración a la pared celular de las plantas causando la pudrición blanda. La capacidad de destruir las plantas se debe a la expresión y secreción de una amplia gama de enzimas hidrolíticas debido a la acción del sistema de inducción de las bacterias Gram-negativas, el cual incluyen celulasas y expansinas. P. carotovorum PC1 FJ890936 se utilizó para el diseño de los cebadores para la amplificación del gen de la celulasa y que incluyera el sitio de exportación celular pelB. El producto de la PCR se clonó en el vector de expresión pET22b-PcExl1 y se transformaron en Escherichia coli. El peso molecular de cada una de las clonas obtenidas fue de 1267 pb. La estimación de los pesos moleculares de la proteína expresada se realizó usando análisis de SDS-PAGE y se obtuvieron dos bandas de 39 kDa y 32 kDa, respectivamente. La actividad enzimática se llevó a cabo por un ensayo de reducción de liberación de azúcares reductores. Sin embargo, la celulasa no mostró una posible interacción con la expansina PcExl1.
