Estandarización de la técnica de rescate de embriones en Alstroemeria

In this research the standardization of Alstroemeria´s embryo rescue technique was performed. Two crossings were made with different varieties of Alstroemeria, from Esmeralda Breeding & Biotechnology (EB&B). The first one was the control crossing which had high germination percentages in vivo. The second one was a crossing with post cigotics barriers that had never given seedlings and in vitro had had less than 2% of germination. Ovule culture and half ovule culture were applied and within the first one the best results in both crossings were achieved. Also, diverse treatments were tried as different sucrose percentages, casein hydrolyzed, glutamine, liquid and solid phase.
En esta investigación se intentó realizar la estandarización del protocolo de rescate de embriones, en el cultivo de Alstroemeria. Se realizó dos cruces diferentes, utilizando variedades de Alstroemeria de Esmeralda Breeding & Biotechnology (EB&B). El primero sirvió de cruce control ya que tiene un alto porcentaje de germinación in vivo y el segundo, es un cruce con barreras de incompatibilidad post cigótica ya que no ha dado semillas y ha tenido menos del 2% de germinación in vitro. Se utilizaron diferentes técnicas de cultivo y diferentes tratamientos con el fin de determinar el medio y la técnica más optima. En las técnicas de cultivo in vitro, se realizó el cultivo de óvulos y el cultivo de óvulos cortados, donde el primero obtuvo los mejores resultados para los dos cruces.
